Tabor, Celia W. (National Institute of Arthritis and Metabolic Diseases, Bethesda, Md.). Stabilization of protoplasts and spheroplasts by spermine and other polyamines. J. Bacteriol. 83:1101-1111. 1962.-Spermine (10(-3)m) or spermidine prevents lysis of lysozyme-produced protoplasts of Escherichia coli W, E. coli B, and Micrococcus lysodeikticus in hypotonic media. Spheroplasts prepared by the action of penicillin are also stabilized by these concentrations of spermine and spermidine, but the protection is not as complete. Streptomycin, polylysine, and Ca(++) are also effective or partially effective stabilizers, but 1,4-diaminobutane, 1,5-diaminopentane, ornithine, Mg(++), and monovalent cations have no protective action at 10(-3)m concentration, and only a slight effect at higher concentrations. The osmotic stability conferred on protoplasts by spermine is irreversible. However, the protective effect of polyamines against lysis is not accompanied by restoration of viability to lysozyme protoplasts. There is a marked reduction in the loss of ultra-violet-absorbing material from the protoplasts to the medium when 10(-3)m spermine is present.

Polyamines inhibit biosynthesis of ethylene in higher plant tissue and fruit protoplasts.
Ethylene production in apple fruit and protoplasts and in leaf tissue was inhibited by spermidine or spermine. These polyamines, as well as putrescine, inhibited auxin-induced ethylene production and the conversion of methionine and 1-aminocyclopropane-1-carboxylic acid to ethylene. Polyamines were more effective as inhibitors of ethylene synthesis at the early, rather than at the late, stages ...
متن کاملChanges of free polyamines in the leaves and stems of ‘Kinnow’ mandarin tree as affected by alternate bearing
In order to investigate the seasonal changes and the possible role of the free polyamines in the leaves and stems on the alternate bearing habit of the ‘Kinnow’ mandarin (Citrus reticulata Blanco) trees, a comparative study was conducted to analyze the levels of free polyamines (putrescine, spermidine and spermine) in the leaves and stems tissues of “on” and &ldquo...
متن کاملThe synthesis of polyamines from methionine in intact and disrupted leaf protoplasts of virus-infected chinese cabbage.
In exploring the role of the chloroplast in the multiplication of turnip yellow mosaic virus, the biosyntheses of the major viral polyamine, spermidine, as well as that of the tetramine, spermine were studied. The synthesis of these polyamines from [2-(14)C]methionine in protoplasts of Chinese cabbage leaf cells derived from healthy plants or those infected by turnip yellow mosaic virus were ex...

An improved analytical method, based on high pressure liquid chromatography, has been developed for the simultaneous determination of the polyamines and S-adenosyl-containing compounds in extracts of plant protoplasts. The method involves simple procedures for sample preparation and permits quantification of 1 picomole or less for all the compounds. This method has been used to study the effect...
متن کاملEvaluation the Effects of Foliar Treatments of Polyamines and Some Organic Acids on Quantitative and Qualitative Traits in Some Pistachio Cultivars
Among the most important problems of pistachio are physiological disorders such as fruit abscission and production of blank nuts. Environmental stresses and inadequate nutrition during flowering time have increased these problems in recent years. In order to evaluate the effects of foliar treatments with polyamines and other compounds (amino acids and ascorbic acid) on quantitative and qualitat...
